WHY ARE LONG INSITU HYBRIDIZATION PROBES BETTER THAN SHORT ONES?

The covalent bond is the strongest atomic bond that exists in nature that has a bond strength of approximately 90 kcal/mol.

 

One of the weakest natural bonds in fluid is known as the hydrogen bond whose bond strength is approximately 1 kcal/mol.

Nucleic acids hybridize between each other by forming hydrogen bonds and for this reason are weak bonds. However long probes make many more bonds than short ones and the sum of many small forces can constitute one great force.

In the case of the HistoSondas some segments are more than 1000 nucleotides long and for this reason the force of the hybridization with its target is very strong.
