INTRODUCTION
Erythroid cells are the only cells that produce hemoglobin. Under normal conditions erythroid cells are located exclusively in adult bone marrow, but during embryonic and fetal periods (as well as in some processes such as myelofibrosis) there is extra medullar hematopoiesis, generally in the spleen and liver.
There are some benign tumors such as myelolipoma of the suprarenal gland and soft tissues, in which ectopic erythropoiesis appears. The erythroid series is relatively difficult to differentiate in normal bone marrow biopsies and even more so in the spleen and liver. It can be very difficult to recognize in biopsies when there is atypical erythropoiesis, such as megaloblastic anemia, Myelodysplastic syndromes and erythroleukemias.
Until now, it has been necessary to use anti-hemoglobin oranti-Glycophorin A antibodies to differentiate the erythroid series. The main problem with these immunohistochemistry techniques is that they also stain erythrocytes, giving a diffuse background that complicates histological observation.
Our probes detect only those cells that synthesize RNA from alpha chain hemoglobin, so only nucleated cells are stained, not counting the scarce eticulocytes (early-stage red cells that still have globin RNA) that may still be in the tissue. Hemoglobin is a molecule in which four globin chains are bound to a HEM group. There are many types of hemoglobins (Adult, fetal, embryonic, etc) but all of them have two alpha globin chains.
INTENDED USE
For use in In Vitro Diagnosis
The Histosonda Hemoglobin Alpha Chain is useful for erythroid cell detection in any tissue, as well as when its morphology is altered by some of the aforementioned pathological processes.
WARNINGS AND PRECAUTIONS
Histosonda Hemoglobin Alpha Chain has been designed for professional use in In Vitro Diagnosis and must be manipulated by qualified and accordingly trained personnel.
In order to obtain the best results, the instructions contained in the manual must be followed. Any change to the indicated temperatures, times or any other step of the process can lead to poor results.
KIT COMPONENTS
The kit includes 20 single test tubes of lyophilized Histosonda Hemoglobin Alpha Chain. Histosonda Hemoglobin Alpha Chain consists of a fragment of single-stranded DNA with a length of 157 nucleotides, complementary to expressed RNA.
